چکیده

Monoclonal antibodies (mAbs) have significantly advanced the treatment of a variety of human diseases, particularly the ability to treat cancer. The therapeutic use of monoclonal antibodies consists in activation of antibody-dependent cell-mediated cytotoxic i ty (ADCC), complement-dependent cytotoxicity (CDC), or engagement with cell surface receptors (to either activate or inhibit signaling) (Figure 1). Currently, most of the antibodies for therapeutic usage are monospecific, with a defined specificity for a particular molecular part of an antigen, known as an epitope. [1] However, despite being the most widely used form of cancer immunotherapy, monoclonal antibodies have not been as successful as expected.

A Comparative Investigation of the Bispecific Antibody: Expression in Expi293F Cells and E.Coli

Aims: Bispecific antibodies are capable of targeting two cell surface markers simultaneously. Blinatumomab an anti-CD3/CD19 Bispecific T cell engager antibody (BiTE) is approved for clinical application by FDA. This BiTE effectively targets malignant cells in ALL (Acute Lymphoblastic Leukemia) patients. For production of large quantities of such antibodies in industrial settings choice of host ...

Woodchuck Hepatitis Virus Post-Transcriptional Regulation Element (WPRE) Promotes Anti-CD19 BiTE Expression in Expi293 Cells

Background: Bispecific antibodies represent an important class of mAbs, with great therapeutic potentials due to their ability to target simultaneously two distinct epitopes. The generation of functional bispecific antibodies with the highest possible yields is particularly critical for the production of these compounds on industrial scales. Anti- CD3 × CD19 bsAb is a bispecific T-cell engager ...

Advances in Bispecific Antibodies Engineering: Novel Concepts for Immunotherapies

Bispecific antibodies are emerging as novel approach for immunotherapies by combining two antigen-recognizing elements into a single construct that is able to simultaneously bind to two distinct targets. Bispecific antibodies can be applied to recruit immunological effector cells for killing of tumor cells or to simultaneously block two signaling pathways or cytokines. Subsequently, they have p...

In Vivo Secretion of Bispecific Antibodies Recruiting Lymphocytic Effector Cells

Engineered Fc-lacking bispecific antibodies have shown an exceptionally high potency for recruiting lymphocyte effector cells and enhancing antitumor activity, which is under evaluation in several clinical trials. However, current treatment regimens raise some issues that should be considered, such as the high cost of clinical-grade bispecific antibodies and the achievement of sustained therape...

Production of Native Bispecific Antibodies in Rabbits

BACKGROUND A natural bispecific antibody, which can be produced by exchanging Fab arms of two IgG4 molecules, was first described in allergic patients receiving therapeutic injections with two distinct allergens. However, no information has been published on the production of natural bispecific antibody in animals. Even more important, establishment of an animal model is a useful approach to in...
